Venous thrombosis occurs at an incidence of 1–2 cases per 1,000 individuals and there are approximately 10 million cases worldwide. In Europe, thrombosis causes approximately 544,000 deaths each year, with a high associated economic cost. There are many reported risk factors, including surgery, pregnancy, use of oral contraceptives and prolonged immobilisation.
Genetic factors also play an important role in developing thrombosis. Factor V Leiden thrombophilia is the most common form of hereditary thrombophilia and occurs in 3.8% of the general US and European heterozygous population. Heterozygotes are 7 times more likely to develop thrombosis, while homozygotes have an 80-fold risk. Prothrombin thrombophilia is the second most common form and occurs in about 1 in 50 people in the US and European Caucasian population. On the other hand, mutations in the MTHFR gene are associated with elevated homocysteine levels, an increased risk of blood clots, miscarriages or neural tube defects in offspring.

The assay can distinguish between individuals who are heterozygous and homozygous for all the above mutations.
